Oral Chronic idiopathic constipation Adult: 24 mcg bid. |

History of mechanical GI obstruction. |
Warnings / Precautions |
Safety has not been established in pregnant women. Not to be used in patients with severe diarrhoea. |
Adverse Reactions |
Dose-dependent nausea. Diarrhoea, abdominal distension, pain, flatulence and vomiting. Headache, dizziness, fatigue, dyspnoea, and peripheral oedema. |
Mechanism of Actions |
Lupristone is a locally-acting chloride-channel activator that increases intestinal secretion. In doing so, it increases intestinal motility, thus aiding the passage of stool and relieve the symptoms associated with chronic idiopathic constipation. Absorption: Absorption after oral admin is negligible. Metabolism: Rapidly and extensively metabolised by carbonyl reductase, likely in the stomach and jejunum. |
