Indications |
Oral Acute psychosis Adult: ≥15 yr: 400-800 mg daily in 2 divided doses. Max: 1.2 g daily. For patients with predominantly negative symptoms: 50-300 mg daily.
Intramuscular Acute psychosis Adult: 400 mg daily.
Special Populations: In renal impairment: Doses to be reduced to 1/2 the normal adult dose for patients with a CrCl (ClCr) of 30-60 mL/min or to 1/3 of the normal adult dose in patients with a ClCr of 10-30 mL/min. |

Pregnancy and lactation, pheochromocytoma, prolactin-dependent tumors. Children <15 yr. | ||||||||||||
Warnings / Precautions |
Renal impairment, Parkinson's disease, CV disease, history of epilepsy, elderly. May affect performance of skilled tasks. withdraw gradually and closely monitored to avoid relapse. Elderly. | ||||||||||||
Adverse Reactions |
Weight gain, dizziness, postural hypotension, extrapyramidal symptoms, neuroleptic malignant syndrome. GI disorders and dry mouth. CNS effects. Hyperprolactinaemia (with galactorrhoea, amenorrhoea, gynecomastia, breast pain, sexual dysfunction). | ||||||||||||
Overdose Reactions |
Symptoms include generalised convulsions, coma, motor restlessness, tachycardia and slight prolongation of the QT interval. | ||||||||||||
Drug Interactions |
Guanethidine and other adrenergic neuron blockers, antiarrhythmics, antihistamines, antimalarials and cisapride, diuretics, general anaesthetics, hypnotics, anxiolytics and opioids. Mechanism of Actions |
Amisulpride is a substituted benzamide atypical antipsychotic with general properties similar to those of sulpiride and is reported to have a high affinity for dopamine D2 and D3 receptors. Absorption: Bioavailability: about 48%. Distribution: Protein binding: about 16%. Metabolism: Limited metabolism. Excretion: Terminal half-life: 12 hr. Mainly excreted unchanged in urine. |
