Tolperisone

Indications
Oral
Spasticity, Muscle spasms
Adult: As hydrochloride: 50-150 mg tid.
Child: As hydrochloride: <6 yr: 5 mg/kg daily in 3 divided doses; 6-14 yr: 2-4 mg/kg daily in 3 divided doses.
Contraindications
Myasthenia gravis.
Adverse Reactions
Muscular weakness, headache, hypotension, nausea, vomiting, abdominal discomfort.
Mechanism of Actions
Tolperisone is a centrally acting muscle relaxant.
Absorption: Well absorbed from GI tract. Peak plasma concentration in 0.5-1 hr. Bioavailability: 20%.
Metabolism: Extensively metabolised by liver and kidney. Elimination half-life (IV): 1.5 hr.
Excretion: Excreted mainly via urine as metabolites.
Administration
Should be taken with food.
